Biotools has exploited a new technology, the “LIONPROBES^® technique” in order to develop sensitive and reliable detection Real Time Amplification Assays. This technology is based on the use of a DNA polymerase with 3´-5´exonuclease proofreading activity (Pfu) and a double labelled oligonucleotide (5´TAMRA—FAM3´) which presents a mismatch at 3´end of the hybridization area. During the amplification, the Pfu recognises the 3´mismatch and excises the mismatched base pairs labelled with the fluorophore (FAM). Once this mismatch is corrected amplification priming is enabled by the oligonucleotide. During amplification cycles the fluorescence emerges as result of the release of the fluorophore.
